چکیده
Objectives: To evaluate the effect of elexacaftor/tezacaftor/ivacaftor in The Netherlands for people with CF (pwCF) 12 years and older at least 1 F508del mutation after 6 months use. Methods: Our study is a prospective observational cohort which we report real life data on use elexacaftor/tezacaftor/ivacaftor. primary endpoint change percent predicted FEV1 (ppFEV1) secondary SCC correlation between ppFEV1 SCC. Results: We enrolled 185,169 (82.8%) homozygous patients 35 (17.2%) heterozygous mutation. baseline mean was 72% 84.4 mmol/L. After there significant increase 13.8% decrease 50.1 There no In subanalysis, female had higher 4.7% compared to men. PwCF who did not another CFTR modulator before, showed 5.5% those that were modulators before. addition, an FEV1>90% FEV1<90%, (non-significant), probably due fact these already used modulator. analysis only significantly different male patients. pwCF decreased 11.8% more group pwCF. Conclusion: conclusion, our show substantial lung function Dutch than age need biomarkers monitor modulators.

RATIONALE Tezacaftor (formerly VX-661) is an investigational small molecule that improves processing and trafficking of the cystic fibrosis transmembrane conductance regulator (CFTR) in vitro, and improves CFTR function alone and in combination with ivacaftor. OBJECTIVES To evaluate the safety and efficacy of tezacaftor monotherapy and of tezacaftor/ivacaftor combination therapy in subjects w...
متن کاملRestoration of CFTR function in patients with cystic fibrosis carrying the F508del-CFTR mutation
Restoration of BECN1/Beclin 1-dependent autophagy and depletion of SQSTM1/p62 by genetic manipulation or autophagy-stimulatory proteostasis regulators, such as cystamine, have positive effects on mouse models of human cystic fibrosis (CF). These measures rescue the functional expression of the most frequent pathogenic CFTR mutant, F508del, at the respiratory epithelial surface and reduce lung i...
متن کاملDecoding F508del Misfolding in Cystic Fibrosis
The functional deficiency of the cystic fibrosis transmembrane conductance regulator (CFTR), a plasma membrane chloride channel, leads to the development of cystic fibrosis. The deletion of a phenylalanine at residue 508 (F508del) is the most common cause of CFTR misfolding leading to the disease. The F508del misfolding originates in the first nucleotide-binding domain (NBD1), which induces a g...

INTRODUCTION Cystic fibrosis (CF) is one of the common genetic disorders in the western world. It has been reported to be very rare in Asian populations. According to the Cystic Fibrosis Genetic Analysis Consortium, more than 1,000 mutations of the CF gene have been identified. The CF gene, named the cystic fibrosis transmembrane conductance regulator (CFTR), is located on chromosome 7 and comp...
